I don't believe so westside, at least I've never heard of it being done.
Unless you are moving tremendous amounts of data on and off your Xbox the HDD should be ok. Or maybe if you are running Linux on one. Remember that most of the data you access on the Xbox (games, apps) is really only being read by the Xbox and not re-written like an OS would do.

No need to defrag. Windows and Linux distros that reside on (I think, have to check) the FS that starts with an R (mind went blank when I was about to type it in lol) are the only ones that need defragging. I think it has to do with the way the FS allows the OS to be written.
To help shed some light, the reason it doesnot need defraging is because as both Jodo and Stray stated the games are read only, the files that are re-written reside in your games saves which are never modified always deleted and saved over, thats how you can delete U an T Data and it comes back with a basic folder set.
Defraging a drive is to put back randomly accessed files, in a location which best suits the OS. a frag drive means the files did not go back to thier original location in order to be speedy. hope that helpssome times I confuse rather then enlighten.
